Virus Isolation in Tissue Culture

 

If there is Cytopathic Effect (CPE) in tissue culture in first passage cultures after 48 hours, and the antigen detection ELISA on either the original material or supernatant from the first passage is positive, then the original sample is positive for the presence of FMDV.

If there is no evidence of CPE after the first passage, a second passage should be performed for a further 48 hours. If there is CPE after the second passage, then the presence of FMDV should be confirmed using the antigen detection ELISA. If no CPE is evident on second passage, and provided the original suspension was negative in the antigen detection ELISA, then the sample should be reported as 'no FMD virus detected'.
